Antigen-induced death of T-lymphocytes.

چکیده
Resting mature T-lymphocytes are activated when they are triggered via their antigen-specific T-cell receptor (TCR) molecule or the associated CD3 antigen. In contrast, preactivated T-cells can undergo activation-induced cell death (AICD) in response to the same signals. Stimulation of activated T-cells upregulates the expression of the Fas-ligand, and the interaction of Fas-ligand with the corresponding Fas receptor triggers an apoptosis program that culminates in cellular suicide usually associated with the fragmentation of DNA into oligonucleosomal bands. Molecular evidence indicates that proteases related to interleukin-1-beta converting enzyme play an essential role in the execution of cell death. AICD of mature T-lymphocytes can be efficiently triggered by monoclonal antibodies against the CD3/TCR complex, or by superantigens such as bacterial enterotoxins. Although it is more difficult to induce AICD by conventional peptide antigens, it is now clear that antigen-induced AICD is a powerful means of eliminating antigen-reactive T-cells. Therefore, AICD contributes to the regulation (i.e., termination) of cellular immune responses. In addition, AICD might play a role in the establishment of peripheral immune tolerance. Increased knowledge of the molecular mechanisms of AICD opens new immunotherapeutical perspectives for the treatment of certain autoimmune diseases, and will have implications in other areas such as transplantation medicine.

T lymphocytes have been characterized to different subsets such as cytotoxic T, Thelper1 (Th1), Th2, Th3, Th9, Th17, and regulatory T cells. Each of these subsets have specific function which distinct them from other lymphocytes. Regulatory T lymphocytes are effective cells in immune system that play an important role in cancers, autoimmune and infectious diseases. Two main subsets of regulator...

Background & Aim: Tolerance to the semi-allogenic fetal graft by the maternal immune system is a medical enigma that has stimulated investigations for a half of century. Several hypotheses have been proposed to explain the tolerance of mother to the fetus. The successful pregnancy is proposed and proved by many scientists to be a Th2 dominant phenomenon. This hypothesis is proved in most as...
متن کاملDelayed Activation-Induced T Lymphocytes Death in Aplastic Anemia: Related with Abnormal Fas System
OBJECTIVES To quantitate apoptosis and Fas antigen expression of T lymphocytes by activation in aplastic anemia (AA) and compare with that of normal controls and completely-recovered AA, and to investigate the apoptotic sensitivity to anti-fas antibody of activated T lymphocytes in AA. METHODS We studied the expression of Fas antigen on fresh T lymphocytes of twenty patients with AA [13 newly...
متن کاملDeath receptor-mediated suicide: a novel target of autoimmune disease treatment.
In the thymus, based on the reactivity of their T-cell receptor with self-MHC and antigenic peptides, developing immature T-cells undergo positive and negative selection. Cells recognising self-peptides and MHC with high affinity are considered autoreactive, and thus potentially harmful, and are eliminated by induction of apoptotic cell death. Thymic negative selection is, however, only incompl...
متن کاملCD152 (CTLA-4) Determines the Unequal Resistance of Th1 and Th2 Cells against Activation-induced Cell Death by a Mechanism Requiring PI3 Kinase Function
Survival of antigen-experienced T cells is essential for the generation of adaptive immune responses. Here, we show that the genetic and antibody-mediated inactivation of CD152 (cytotoxic T lymphocyte antigen 4) in T helper (Th) effector cells reduced the frequency of nonapoptotic cells in a completely Fas/Fas ligand (FasL)-dependent manner. CD152 cross-linking together with stimulation of CD3 ...
